چکیده
Objective: To investigate the effects of matrine on the proliferation and apoptosis of RPMI 8226 cells in vitro. Methods: RPMI 8226 cells were cultured in vitro, in the presence of different concentrations of matrine. The effect of matrine at different concentrations on the proliferation and apoptotic rate of RPMI 8226 cells was detected by MTT assay and flow cytometry. The mRNA expression of cyclooxygenase-2 (COX-2) was determined by reverse transcription polymerase chain reaction (RT-PCR) while the expression level of COX-2 protein was evaluated by Western blotting. Results: Matrine inhibits the proliferation and induces apoptosis of RPMI 8226 cells in a timeand concentration-dependent manner. In addition, it induced COX-2 mRNA expression and decreased COX-2 protein expression in RPMI 8226 cells. Conclusion: Matrine can inhibit the proliferation and induce apoptosis of RPMI 8226 cells possibly via the downregulation of COX-2 gene as one of the mechanisms.

A preliminary study of the effect of curcumin on the expression of p53 protein in a human multiple myeloma cell line
Curcumin is an inexpensive, natural plant ingredient with protease inhibitor effects. The present study aimed to analyze the inhibitory effects of curcumin on the multiple myeloma (MM) RPMI 8226 cell line, and examine the underlying mechanism that promotes the apoptosis of RPMI 8226 cells. A growth curve was constructed in order to observe the relative growth velocity, and MTT was used to analy...
متن کاملMatrine inhibits diethylnitrosamine-induced HCC proliferation in rats through inducing apoptosis via p53, Bax-dependent caspase-3 activation pathway and down-regulating MLCK overexpression
The proliferation of hepatocellular carcinoma (HCC) cells is one of the leading causes of liver cancer mortality in humans. The inhibiting effects of matrine on HCC cell proliferation have been studied, but the mechanism of that inhibition has not been fully elucidated. Since, apoptosis plays an important role in HCC cell proliferation. We examined the apoptosis-inducing effect of matrine on tu...

متن کاملEvaluation of in vitro effects of various targeted drugs on plasma cells and putative neoplastic stem cells in patients with multiple myeloma
Multiple myeloma (MM) is a malignancy characterized by monoclonal paraproteinemia and tissue plasmocytosis. In advanced MM cytopenia and osteopathy may occur. Although several effective treatment strategies have been developed in recent years, there is still a need to identify new drug targets and to develop more effective therapies for patients with advanced MM. We examined the effects of 15 t...
